More Road and River Trivia (I am beating a dead horse):
It looks like there was a bypass to the Ohio River at about the end of the 1790's. I have seen some accounts that Gist's trace was based on earlier Indian Trails.

That site has led to a wealth of others, but the article below may cover the original route. Looks like the author has a book on the broader subject of trails.
I think Wayne covered Mr. Knee's question with respect to water routes and land routes, but this may add some additional information about where the barges were launched.
One confusing term is the use of Cumberland on two distinct roads. The "Cumberland Trail" and the "Cumberland Gap" are hundreds of miles appart. I believe Cumberland, Maryland was at about the point where Maryland narrows between Pennsylvania and Virginia. (See also Bradoock's Road, National Road, Route 40, etc. in earlier posts.)
